Cloning and functional research of miRNA (Bra-miR07) associated with regulation of early organ development in cauliflower 03-25-2016

The overexpression of Bra-miR07 can significantly influence the growth and development of transgenic plants and Bra-miR07 may also play important roles in regulation of cauliflower organ development.

 

In the previous researches, researchers obtained a series of new miRNAs that showed significantly different expression in the early organ development of cauliflower by high-throughput sequencing. While in the present research, researchers Zhang Qingli, Jin Chuan, Qin Erjun, Wang Yu Wu Mei, Li Lihong, Li Hui, Chen Chengbin, Song Wenqin and Wang Chunguo further analyzed a new miRNA (named Bra-miR07) that showed different expression between cauliflower hypocotyl and cotyledon.


The results indicate that the sequence length of Bra-miR07 precursor is 270 bp which can form the stable secondary hairpin structure. Bra-miR07 appears higher expression in cauliflower cotyledon than that in hypocotyl. To further uncover the function of Bra-miR07, researchers construct and transfer overexpression vector of Bra-miR07 into Arabidopsis thaliana by floral dip, and successfully obtain the Bra-miR07 overexpression transgenic plants. Phenotypic analysis shows that overexpression of Bra-miR07 can significantly influence the growth and development of transgenic plants. Compared with wild-type Arabidopsis, Bra-miR07 transgenic plants show slow growth, dwarf plant and lower fertility. Consequently, these results imply that Bra-miR07 may also play important roles in regulation of cauliflower organ development.
